    研究方向

    主要研究方向:炎症相关疾病的新靶点和新药发现研究研究背景:炎症是机体对损伤因子所发生的防御反应。然而,作为一柄双刃剑,过度或失控的炎症反应(天然免疫系统过度激活和炎症细胞因子过量表达)会导致组织损伤介导多种疾病的发生发展,是一种常见的病理过程,在多种疾病,如脓毒血症、肥胖与糖尿病及其并发症等代谢性疾病中起着重要的介导作用。因此,抑制过度炎症的药物有望应用于多种炎症相关性疾病的治疗。代谢和心血管疾病已成为人民健康的重要威胁,研发代谢相关疾病防治的新策略和新手段具有重要的医学价值与社会意义。慢性炎症是多种代谢性疾病的重要病理过程。在代谢性疾病的发生发展过程中,初始代谢异常的免疫应答导致机体和局部过度炎症的发生,过度炎症又导致病灶产生更多的炎性浸润放大炎症反应,进一步直接损伤组织细胞或代谢信号通路,它们正反馈式地促进代谢性疾病的发生发展。因此,抑制过度炎症反应可能成为多种代谢性疾病的治疗新策略。科学问题:虽然过度炎症与代谢性疾病的关系已有大量报道,但是,(1)新机制和新靶点:代谢紊乱的刺激物(高血糖、高血脂、高血压等)如何激活炎症反应?是否能从慢性炎症发生的过程和炎症调控的信号通路中找到防治代谢性疾病的新靶点?(2)抗炎新化合物:目前临床上的甾体或非甾体类抗炎药均靶向环氧化酶/前列腺素系统和炎性血管反应,并非靶向炎症因子过量表达的炎症信号通路,靶标单一,局限性强,无法用于慢性炎性疾病的治疗;而针对炎症因子及其受体的抗体类药物,其靶点在炎症发生的下游,对炎症因子的抑制种类单一,对大量多种炎症因子复合的炎症相关疾病和代谢性疾病治疗效果有限。因此,如何去寻找抑制过度炎症反应的新型抑制剂和候选新药?(3)候选新药:如何确证抑制过度炎症反应可以成为治疗代谢相关疾病的重要手段?结合新靶点和新化合物,能否寻找到基于抗炎机制治疗代谢性疾病和急性炎症疾病的候选新药?研究内容:我们瞄准过度炎症这一重要病理过程,以代谢性疾病和脓毒血症防治为目标导向,综合运用药理学、药物化学、分子生物学等多学科手段,主要在代谢性疾病中的慢性炎症发生机制和调控机制、抗炎新药发现和靶点确证、代谢性疾病的抗炎防治和新药发现等三个方面取得了系列研究成果,为代谢性疾病的抗炎治疗提供了新的干预策略和候选新药。(1) 代谢性疾病中的慢性炎症发生机制和调控机制:已完成的主要研究包括:首次发现饱和脂肪酸直接结合细胞膜上的MD2蛋白而激活炎症信号通路和心脏炎性损伤(Nat Comm, 2017);阐明了血管紧张素通过促炎导致心肌损伤的新机制和靶点(Hypertension,2015等);发现JNK2介导了糖尿病心肌病和肾病中的过度炎症发生过程(Diabetes, 2014;Kidney Int, 2017);提出MD2、JNK2等可以成为治疗糖尿病并发症的有效靶点。目前正在开展研究包括:高血糖激活心肌细胞炎症反应的分子机制;动脉粥样硬化中炎症反应的发生机制;炎症发生的重要调控蛋白MyD88在代谢性心血管疾病中的作用和机制;寻找调控炎症的新型蛋白激酶及其在糖尿病心肾并发症中的作用和机制;寻找调控炎症的新型泛素化和去泛素化酶及其在急慢性炎症疾病中的作用与机制。(2) 抗炎新化合物发现和靶点确证:已完成的主要研究包括:基于天然先导和靶点结构进行合理药物设计,合成了千余个小分子化合物,并利用“LPS-巨噬细胞-炎症因子释放”体系,从中筛选发现了50个新型抗炎活性化合物(J Med Chem, 九篇);通过药理机制研究,阐明了部分活性化合物抗炎的直接分子靶点,如MD2和JNK等(BJP, 六篇)。目前正在开展研究包括:含氮杂环类化合物中设计和寻找新型抗炎化合物;以天然产物为先导物的药物化学结构优化与抗炎新化合物设计发现;天然产物中的抗炎化合物的分子靶点和化学生物学研究;靶向MD2、MyD88、DCLK1、PLCy等蛋白的新型抗炎化合物设计合成与评估;新型抗炎短肽的设计与发现;新型抗炎药物设计和靶点分子动力学模拟研究。(3)代谢性疾病的抗炎防治和新药发现:  将自己发现的多个抗炎活性化合物用于疾病治疗,阐明了它们的药效作用和药理机制,并对多个新化合物进行了成药性评价。其中,靶向MD2的新化合物X22作为脓毒血症的1.1类候选新药,靶向JNK2的新化合物C66作为糖尿病肾病的1.1类候选新药,都已进入临床前研发阶段,并获得国家新药创制重大专项支持。目前正在开展其他几个候选新化合物的成药性评价和新药临床前研究,它们基于抗炎靶点和机制有望治疗糖尿病肾病、高血压心衰和结肠炎。其它研究方向:抗肿瘤药物化学与抗肿瘤药理机制研究1 抗肿瘤药物化学研究1.1 以天然产物为先导物的药物设计、合成、抗肿瘤构效关系研究1.2 靶向IKK、EGFR、FGFRs等靶点的抗肿瘤药物设计、合成和构效关系研究1.3 新型抗肿瘤药物设计和靶点分子动力学模拟研究2 抗肿瘤药理机制研究2.1新发现的抗肿瘤活性化合物的药理机制和药效研究2.2 天然产物及其类似物通过激活ROS诱导肿瘤凋亡的作用机制研究2.3旁路激活与肿瘤耐药的分子机制研究2.4 端粒与G4在炎癌转化的作用机制研究2.5 肿瘤中lncRNA和miRNA的作用和机制研究
